چکیده
UNLABELLED PREMISE OF THE STUDY Primers for 31 microsatellite-containing loci were developed for the threatened orchid Platanthera praeclara to enable characterization of the population genetics of this tallgrass prairie native. • METHODS AND RESULTS Sixteen polymorphic microsatellite loci were identified from four populations. Six of these loci were not in linkage disequilibrium. The average number of alleles per locus per population ranged from 6.4 to 8.9. • CONCLUSIONS The results indicate that six of the polymorphic loci will be useful in future studies of population structure, gene flow, and genetic diversity.

Spatial and temporal optimization in habitat placement for a threatened plant: the case of the western prairie fringed orchid
This paper investigates an optimization approach to determining the placement and timing of habitat protection for the western prairie fringed orchid. This plant’s population dynamics are complex, creating a challenging optimization problem. The sensitivity of the orchid to random climate conditions is handled probabilistically. The plant’s seed, protocorm and above-ground stages are all accoun...
متن کاملCharacterization of microsatellite primers in the endangered orchid Phaius australis and cross-amplification to P. bernaysii (Orchidaceae)1
Premise of the study The swamp orchid, Phaius australis (Orchidaceae), is nationally endangered due to illegal collection and habitat loss and fragmentation, resulting in a disjunct distribution in spring and coastal wetland ecotones along Australia's east coast. Polymorphic microsatellite markers were developed to study genetic diversity and population structure for conservation and restoratio...

PREMISE OF THE STUDY Twenty-six microsatellite markers were developed for the endangered orchid Cypripedium japonicum (Orchidaceae) to estimate the clonal diversity and genetic structure of the remaining populations in Japan. METHODS AND RESULTS Microsatellite loci of C. japonicum were isolated using Ion Personal Genome Machine (PGM) sequencing. The primer sets were tested on 55 ramets sample...
متن کاملMycorrhizal Fungal Diversity and Community Composition in Two Closely Related Platanthera (Orchidaceae) Species
While it is generally acknowledged that orchid species rely on mycorrhizal fungi for completion of their life cycle, little is yet known about how mycorrhizal fungal diversity and community composition vary within and between closely related orchid taxa. In this study, we used 454 amplicon pyrosequencing to investigate variation in mycorrhizal communities between pure (allopatric) and mixed (sy...
متن کاملMolecular Evolution of Chloroplast Genomes of Orchid Species: Insights into Phylogenetic Relationship and Adaptive Evolution
Orchidaceae is the 3rd largest family of angiosperms, an evolved young branch of monocotyledons. This family contains a number of economically-important horticulture and flowering plants. However, the limited availability of genomic information largely hindered the study of molecular evolution and phylogeny of Orchidaceae. In this study, we determined the evolutionary characteristics of whole c...
